The QIAsymphony DNA Investigator Kit enables automated purification of genomic DNA from 1–96 samples, such as swabs, filters, casework or crime-scene samples, and blood on the QIAsymphony SP. Purification is fast and efficient, and purified DNA performs well in downstream analyses, such as quantitative PCR and STR analysis, with high signal-to-noise ratios.

The QIAsymphony DNA Investigator Kit enables highly efficient purification of DNA, even from very small amounts of sample material. The combination of sample input volumes of up to 1000 µl with elution volumes as low as 30 µl enables highly sensitive detection of DNA from a wide range of sample types, including diffuse stains.


The QIAsymphony DNA Investigator Kit combines the speed and efficiency of silica-based purification of genomic and mitochondrial DNA with the convenient handling of magnetic particles. Innovative, ready-to-run reagent cartridges are prefilled with magnetic particles and all reagents required for the purification procedure and are automatically opened by the QIAsymphony SP, which helps to minimize the risk of environmental contamination. Worktable setup is rapid, which saves your valuable time. Simply place up to 2 reagent cartridges in the "Reagents and Consumables" drawer. The reagent cartridges can be either from the same kit or from different kits, enabling different purification procedures to be performed within the same run of 96 samples. Reagent volumes only for the selected number of samples are used, giving you complete cost control. For increased flexibility, DNA can be eluted in volumes of between 30 and 400 µl of water or Buffer ATE (modified TE buffer).


The purification procedure is designed to ensure safe and reproducible handling of precious or potentially infectious samples, and comprises 4 steps: lyse, bind, wash and elute. The user can choose different elution volumes between 30 ?l to 400 ?l of water or modified TE Buffer (Buffer ATE), depending on the protocol.

Since the type of samples that can be processed using the QIAsymphony DNA Investigator Kit can vary greatly, there is also a variety of different pretreatments, optimized for specific sample types. Samples are lysed under denaturing conditions in the presence of proteinase K and Buffer ATL in total volumes of either 200 ?l, 500 ?l or 1000 ?l.

There are two different kinds of DNA purification protocols, which can be used in conjunction with the pretreatment protocols. Casework protocols purify genomic and mitochondrial DNA from 200 ?l, 500 ?l or 1000 ?l of lysate obtained from the sample lysis procedures detailed in the corresponding pretreatment protocols. DNA can be eluted in 30–200 ?l of either water or modified TE Buffer (Buffer ATE). Reference protocols purify DNA from database samples, such as buccal swabs or dried blood. DNA can be eluted in 100–400 ?l of Buffer ATE. The purified DNA is ready to use in downstream applications.
