Performance

The DNeasy mericon 96 QIAcube HT Kit is designed for rapid, automated purification of DNA from a variety of raw and processed food matrices, while minimizing the carryover of PCR inhibitors inherent to complex food samples. DNeasy mericon purification kits provide a universally applicable extraction method that gives reliable results even when using strongly inhibitory, highly processed, fatty, acidic, high-, or low-DNA content foods. The QIAcube HT platform, together with the dedicated DNeasy mericon 96 QIAcube HT Kit, enables automated mid- to high-throughput DNA purification from food sample types. Now it is possible to prepare the same high-quality DNA as from the manual DNeasy mericon Food Kit with a quick and simple automated procedure.

Principle

The DNeasy mericon 96 QIAcube HT Kit enables automated purification of nucleic acids from up to 300 mg food sample material using the QIAcube HT instrument. The procedure yields high-quality DNA that performs well in PCR and other downstream applications.

The DNeasy mericon 96 QIAcube HT Kit combines the selective binding properties of a silica-based membrane with a high-throughput 96-well format, and is designed for fully automated, simultaneous processing of 24–96 samples on the QIAcube HT instrument.

Procedure

The DNeasy mericon 96 QIAcube HT Kit uses a modified cetyltrimethylammonium bromide (CTAB) extraction procedure for rapid automated purification of DNA from food matrices on the QIAcube HT instrument. Homogenized food samples undergo a manual pretreatment step for efficient lysis using Food Lysis Buffer in combination with proteinase K. Chloroform extraction removes any CTAB-protein, CTAB-debris, or CTAB-polysaccharide complexes. The aqueous phase containing the DNA is transferred to a 96-well S-Block, and all subsequent  purification steps are automated on the QIAcube HT instrument. 

QIAcube HT adds Buffer PB to the aqueous phase and transfers the mixture to the DNeasy 96 plate. As vacuum is applied, nucleic acids are adsorbed onto the silica membrane while contaminants pass through. Washing removes any remaining contaminants and inhibitors, and the DNA is eluted in Buffer EB. The resulting purified DNA is ready for use in a downstream mericon real-time PCR assay.
